Will I lose ride comfort or get additional noise if I use Cusco pillowball mounts?

I assume you're referring to the strut pillow ball tops?
Altho anything in a pillowball configuration will carry increased road noise. But the tradeoff is less flex and more accuracy in handling dynamics.

You assumed right
hmm....
Noise and harshness? OR just noise?

Less movement, means less takeup from a more 'flexible' component, so the trade off is generally noise and less premium ride quality which can be related to harshness. Many factors take place to make a ride all out Harsh.
